Over the course of Avengers: Infinity War and Endgame, Thanos did massive damage to Earth's Mightiest Heroes through his Infinity Gauntlet. Several characters important to the Marvel Cinematic Universe are dead and not coming back. Although Tony Stark sacrificed himself to destroy the Mad Titan, Thanos will still definitely do damage to the heroes who defeated him going forward.

It's totally possible the Thanos from another multiverse might make his way to the MCU in the future, though that's probably unlikely. Instead, we know Thanos keeps a lot of his plans hidden. All the films in the MCU so far have proven Thanos doesn't tend to work alone, so it's possible one of his allies might end up returning to wreak havoc. There's also one Black Order member from the comics missing in the movies: Supergiant, who might end up playing a role going forward.

Although Thanos is dead, there's a chance he might appear again in the future, as The Eternals film is rumored to be a sequel, and it may feature Eros of Titan, who is the brother of Thanos.
